Many local residents have opposed plans for it, which include a large amount of housing.
Ms Blears visited the school recently and met worried residents.
Mr Mulholland is asking that she makes sure that the Government does not overturn the community's wishes by allowing the proposed development to take place. He said: "It's vital that a government inspector does not come along and make a decision that runs contrary to wishes of local residents.
"I hope that Hazel Blears, having spoken to local people, will make sure that Government officials back local residents in this dispute.
The plans are completely unacceptable and local residents are understandably angry that school has refused to listen to their views."
The Girls High School has merged with Leeds Grammar School at its site in Alwoodley.
